BIO

Our research investigates how a soil bacterium, Agrobacterium tumefaciens , genetically engineers plants. Agrobacterium transfers a piece of bacterial DNA, the T-(transferred) DNA, to wounded plant cells where it makes its way through the cytoplasm to the nucleus. Once in the nucleus, T-DNA integrates into the host genome and expresses genes. Under normal circumstances, these genes cause the disease Crown Gall on plants. However, scientists have learned to manipulate T-DNA, replacing disease genes with genes of benefit to the plant. Many genetically engineered crop plants with desirable traits (disease resistance, herbicide tolerance, and enhanced nutritional value) were generated using Agrobacterium . Unfortunately, many important crop plants, including those important to Indiana farmers (corn, soybeans, and wheat) remain highly recalcitrant to Agrobacterium -mediated genetic transformation.

Our research focuses on understanding the role of plant genes and proteins in this natural genetic engineering process. We have identified plant genes involved in bacterial attachment to plant cells, T-DNA and Virulence transfer to and cytoplasmic trafficking within plants , T-DNA nuclear targeting, and T-DNA integration. Recently, we have been able to manipulate some of these genes to improve Agrobacterium transformation efficiency. We are currently working with agricultural biotechnology companies to improve the genetic engineering of crops, including those important for Indianas economy.

  • November 15-17, 2015. Stanton Gelvin attended a NSF-sponsored workshop on plant transformation at Clearwater Beach, Florida.
  • July 17-21, 2016. Stanton Gelvin presented a talk at the 2016 international IS-MPMI (International Society for Molecular Plant-Microbe Interactions) conference at a special session on “Recent advances Agrobacterium biology” entitled “Potential novel role for the Agrobacterium virulence effector protein VirE2 in modulating plant gene expression”. Work from his laboratory was also featured in a presentation by Kiran S. Mysore entitled “Activity of plant non-homologous end-joining DNA repair proteins is not required for Agrobacterium T-DNA integration”.
  • August 15-16, 2016. Stanton Gelvin presented the Keynote Address at the 33 rd Annual Mid-Atlantic Plant Molecular Biology Society meeting at Patuxent National Wildlife Center, Maryland. The talk was entitled “Plant genes important for Agrobacterium -mediated genetic transformation”.
  • September 9-20, 2016. Stanton Gelvin attended the first Euro-Mediterranean Agrobacterium Meeting in Gif-sur-Yvette, France. He chaired a session, and presented an invited talk entitled "New insights into the mechanism of Agrobacterium T-DNA integration into the plant genome." Other members of the Gelvin laboratory also presented invited talks: Lan-Ying Lee "VIP1 and its family members are not important for Agrobacterium-mediated transformation", and Rachelle Lapham "Potential novel role for the Agrobacterium virulence effector protein VirE2 in modulating plant gene expression". Drs. Gelvin and Lee also visited colleagues in Strasboug, France and Basel, Switzerland.
  • October 17-19, 2016. Stanton Gelvin and Lan-Ying Lee attended the 2016 International Symposium on Plant Transformation Biotechnologies at the Academia Sinica, Taipei, Taiwan. Dr. Gelvin presented a talk entitled "Higher order Arabidopsis DNA repair and recombination mutants integrate increasing amounts of Agrobacterium T-DNA into the plant genome." Dr. Lee presented a talk entitled "VIP1 and its family members are not important for Agrobacterium-mediated transformation." Drs. Gelvin and Lee also consulted with colleagues at the Academia Sinica.

  • June 13-14, 2018. Professor Gelvin and Research Scientist Dr. Lan-Ying Lee participated in an Advisory Committee meeting for SweTree Technologies, Umea, Sweden.
  • August 19-23, 2018: Professor Stanton Gelvin and Research Scientist Dr. Lan-Ying Lee attended the International Association of Plant Biotechnology (IAPB) congress in Dublin, Ireland. Dr. Gelvin presented a Keynote talk entitled “Mutation of a Plant Myb Transcription Factor Increases Susceptibility to Agrobacterium -mediated Transformation”. Dr. Lee presented a poster entitled “Generation of Agrobacterium Strains that Efficiently Introduce but Do Not Integrate T-DNA into the Plant Genome”. This poster won an award for best poster of the meeting.

  • September 12-13, 2018: Prof. Stanton Gelvin, Research Scientist Dr. Lan-Ying Lee, and graduate student Rachelle Lapham attended the Agrobacterium 2018 conference at Ghent, Belgium. Dr. Lee presented a talk and poster entitled “Directing VirE2 to the host nucleus abolishes its function in transformation”.   • December 15-23, 2018: Prof. Stanton Gelvin visited the Rubber Tree Research Institute, Chinese Academy of Tropical Agricultural Sciences, Heikou, Hainan, China where he presented three seminars entitled "Plant genetic engineering using Agrobacterium -mediated transformation"; "A plant myb transcription factor negatively regulates Agrobacterium-mediated transformation: Application to crop transformation"; and "Mechanism and control of T-DNA integration into the plant genome".
